This dataset comprises the mouse clonal lineage-tracing and single-cell transcriptomic component of the study entitled “Disruption of cell-type-specific molecular programs of striatal spiny projection neurons in autism.” STICR clonal barcoding combined with 10x Genomics single-cell RNA-seq (3′ v3.1) of virally labeled cells from the cortex, striatum, hippocampus, thalamus, and olfactory bulb of postnatal day 4 16p11.2 deletion (16p11.2df) and wild-type mice. Neural progenitors were labeled at E12.5 by in utero injection of the STICR lentiviral barcode library, and labeled cells were isolated by FACS for sequencing. The dataset includes transcriptome and STICR lineage barcode libraries from 14 brains (9 wild-type, 5 16p11.2df), comprising 269,499 high-quality cells with clonal barcodes. These data were used to compare clonal lineage relationships, cell-type composition, and gene expression between genotypes, and support Yuan et al., “Disruption of cell-type-specific molecular programs of striatal spiny projection neurons in autism.”
