Human Neocortical Glutamatergic Neurons Revealed Through Multimodal Profiling
The human neocortex underlies higher cognition, yet our understanding of its neuronal diversity is limited by sparse access to viable tissue, inconsistent cell type definitions and limited multimodal data. Although single-cell transcriptomics has transformed our understanding of cell type diversity, transcriptomic data alone cannot explain neuronal function. Here, we introduce a multimodal Patch-seq resource of human neocortical glutamatergic neurons spanning cortical layers 2-6. Combining morphoelectric features with spatial transcriptomic data to provide laminar context, we sampled 39 of 42 transcriptomically-defined glutamatergic neuron types. This systematic approach enabled identification of unique phenotypes for understudied populations in human neocortex including, L4 intratelencephalic, L5/6 near projecting and diverse L6 neurons. Human, macaque and mouse comparisons of transcriptomically homologous L5 neurons reveal that morphoelectric differences across species were subclass specific. This dataset provides a unified multimodal reference for studying human cortical circuitry and neuronal diversity.
